Can waking from sleep issues be caused by continuing to run my OC from a USB drive instead of installing it to EFI on the primary drive?

I'm still tweaking things before I do that final step. Currently I'm working through the display off upon waking problem I described in this post.

Could all of the kexts and whatnot running from USB be causing the problem?
